Suppose mtu size mismatche then what will happen with layer 3 protocol neighbourship will be breack or not?

It depends upon that which layer 3 protocol is configured on your routers. If EIGRP is configured, then neighbourship not break, if the MTU is mismatched. But in case of OSPF, MTU mismatch will not allowed to form neighbourship between two routers. But we can run the interface subcommand ip ospf ignore-mtu to ignore MTU check.
